In running is it difficulty>distance as in like running only 1 mile with a weighted vest on or is it distance>difficulty and in running three mile no weighted vest

Obviously depends on the amount of weight lol
 
I want to lose weight and gain muscle simultaneously
Then no running. Because adding weight add pressure on joints & spine while running.

Instead, use a weighted vest while doing incline speed walk. The incline works the legs, increases the heart rate and calories expenditure, while minimizing joints/spine pressure
Also you can use "Heavyhands" method to increase calories expenditure
