Step 5: Deprotection of Amino Acids
After each amino acid is added, the protecting groups on the amino acid side chains are removed (deprotected) using an appropriate reagent, such as trifluoroacetic acid (TFA) or another deprotecting agent. This allows the next amino acid to be added to the chain.
Step 6: Cleavage from the Resin
Once the entire sequence of 191 amino acids is added, the peptide is cleaved from the resin. This is typically done using a cleavage reagent like TFA, which also removes any remaining protecting groups from the peptide chain.
Step 7: Purification
The synthesized growth hormone is purified using techniques such as high-performance liquid chromatography (HPLC), ion exchange chromatography, or reverse-phase chromatography. These methods help to remove any byproducts, unreacted amino acids, and other impurities.
Step 8: Final Refinement and Quality Control
The purified growth hormone is further refined and analyzed using mass spectrometry (MS) or nuclear magnetic resonance (NMR) to confirm its structure and purity. This step ensures the hormone is biologically active and free of contaminants.
Step 9: Formulation and Storage
