Introduction

In this Info-dump I will go over PTH1R (Parathyroid hormone 1 receptor) Analogues and I will explain what they do.


What are PTH1R Analogues?

PTH1R Analogues are synthetic versions of the parathyroid hormone (PTH) which bind to Parathyroid hormone 1 receptors, in turn this increases cAMP (Cyclic adenosine monophosphate) via adenylate cyclase stimulation, which activates PKA (Protein Kinase A) which results in glycogenolysis regulation, PKA also influences both sugar and lipid metabolism, PTH1Rs also assist in regulation of calcium and phosphate levels.


What are osteoblasts?
osteoblasts are cells responsible for mineralization of bone tissue, PTH1Rs promote osteoblasts if the osteoblasts are proliferating via ERK1/2 phosphorylation, in differentiated osteoblastic cells though it tends to do the opposite and actually inhibits ERK1/2

So PTH1Rs can increase bone mineral density(BMD) If Osteoblasts are proliferating.

Abaloparatide and Teriparatide

Abaloparatide and Teriparatide are both in the family of PTH1R agonists, I will be comparing both via studies, thankfully This study includes charts to give some information.

This study shows that Teriparatide shows an increase in BMD from baseline, with an 8.6% rise in the Lumbar Spine (LS) and a 3.6% increase in total hip (TH) compared to placebo

As for Abaloparatide it shows an increase too, it has a 10.4% increase in BMD in Lumar Spine(LS) and a 4.3% increase in Total Hip (TH) which means it has 1.8% more of a BMD increase in LS and a 0.7% BMD increase in TH compared to teriparatide if you compare both daily doses, unlike teriparatide though Abaloparatide shows a selective preference for the G protein dependent (GTPyS sensitive) receptor which results in a reduced duration of intracellular activity, reduced calcium response and lower RANKL production, RANKL (Receptor Activator of Nuclear Factor Kappa-B Ligand)promotes differentiation and activation of osteoclasts, osteoclasts are bone cells which remove old or damaged bone tissue during bone remodeling.

Reconstitution
To reconstitute abaloparatide, you will need an alcohol swab, BAC (bacteriostatic) water, and an insulin needle, it works the same as any other peptide, theres a looksmax.org thread on how to reconstitute peptides so i'll just link it below, although it is pretty water information.
