we know that coffee descends you,
- the high production of cortisol tolls on the skin
- vasoconstriction that affects misc
- over activation of the facial muscles
- restricts water from moisturizing the skin
but how does that even happen?
we are going to be looking at the over-activity of caMKII in the heart tissue, a direct effect of coffee
WHATS CAMKII?

Ca2+/calmodulin-dependent protein kinase II is the larger name for it, which means an enzyme powered by calcium
- its like a machine, which can be used to tag phosphates to another structure creating permanent changes

there exists p-CaMKII in the brain, but what does it look like in the heart?

It was recently shown that CaMKIIδ activation promotes its subcellular mobility, which may facilitate phosphorylation of downstream targets outside the dyadic cleft to regulate contractile function, electrophysiology, and gene transcription.<a href="https://www.ahajournals.org/doi/10.1161/circresaha.120.318402#core-collateral-R27" data-xml-rid="R27">27</a> Figure 2E shows fluorescence recovery after photobleaching experiments in cardiomyocytes expressing GFP-tagged WT or S280A CaMKIIδ. High-glucose treatment increased the fluorescence recovery after photobleaching kinetics for WT but not for CaMKIIδ-S280A (Figure 2E), indicating that this increased CaMKIIδ mobility is also dependent upon S280 O-GlcNAcylation."
all that says is that caMKII activation shows muscle contraction in certain parts of the heart, which was discovered by photobleaching

THIS GRAPH LOOKS COMPLICATED BUT HERES WHAT ITS TELLING US!

Do you see those mountain graphs? it compared contraction of hearts through CaMKII after given different mL/dL amounts of glucose
- The graphs next to it are heart rates

Aight so thats the bigger photo, glucose has direct influence on CaMKII domain opening

I compared the heart rate plots of CaMKII and other activated enzyme, as you can see CaMKII's plots are relatively stable which can tell us the links between heart contraction and CaMKII activity are incredibly strong.

These are two representations of heart beat across a horizontal line, it's the progression of heart beats after given caffiene
The heart beat on the right is spread, showing a steep decline in CaMKII activation after given caffiene
The other one is very tight and consistent, showing how caffeine tightly influences the caMKII to produce

Downstream Consequences in Cardiac Tissue

  • Arrhythmogenesis: Activated CaMKII phosphorylates RyR2 and phospholamban (PLN). This creates a positive feedback loop—further increasing SR calcium leak during diastole, which can trigger delayed afterdepolarizations (DADs) and cardiac arrhythmias.
  • Do you remember that spread heart beat up there? Its a very odd pattern to have, but that's how caffeine influence CaMKII

Oh reminded me to work on my own thread😢
