For those unfamiliar Yoda1 is a piezo1 agonist, it functions by directly activating the mechanosensitive Piezo1 ion channel. This specific, chemically-induced calcium surge is what kicks off the cellular signal to rebuild bone tissue, perfectly bypassing the requirement for physical impact or exercise.

EGF binds EGFR, activating Rac1 for actin rearrangement. This creates membrane ruffles that form a macropinosome. Simultaneously, Yoda1 opens Piezo1, causing Ca^2+ influx. This calcium surge directly activates KCa3.1 channels. Alternatively, SKA-31 activates KCa3.1, while TRAM-34 blocks it. Open KCa3.1 channels heavily suppress Rac1 activity. This blockade halts actin remodeling, stopping macropinosome creation.

How does is it functionally different from HGH?
HGH is an anabolic peptide hormone, it functions by directly stimulating systemic and local IGF-1 production. This specific, growth-factor-induced phosphorylation cascade is what kicks off the cellular signal to rebuild bone tissue, perfectly driving skeletal elongation and mineralization throughout the body.

HGH binds its receptor, stimulating hepatic IGF-1 production. This triggers downstream differentiation, proliferation, and growth. Simultaneously, IL-6 activates the gp130/JAK2 complex. This inflammatory signal attempts to drive STAT3 nuclear translocation. Alternatively, HGH upregulates SHP-2 and SOCS-3 inside the cytosol. Elevated SHP-2 and SOCS-3 heavily suppress JAK2 activation. This structural blockade halts STAT3 signaling, stopping inflammatory gene transcription.

HGH and Yoda1 use different pathways but essentially do the same thing, if you have any questions feel free to ask me.
