1. What is GHRP2?
It’s a growth hormone secretagogue (GHS) similar to GHRP6 and Ipamorelin. However, it provides stronger GH response than these two and has less side effects. It’s also a ghrelin (the hormone that stimulates GH secretion and appetite) receptor agonist.

2. How does GHRP2 work?
As a ghrelin receptor agonist, it binds to your GHSRs (growth hormone secretagogue receptors) to make you hungrier and promote GH release. If we want to get specific, it binds to receptor type 1a (GHS-R1A), which is the particular receptor type that responds to ghrelin. Its appetite stimulation is moderate in comparison to other GHS variants. After binding to your GHSR (specifically GHS-R1A), it creates a signaling chain within the cell that increases calcium levels, which signals the cell to release GH quicker.

TLDR: GHRP2 binds to your receptors to make you hungrier and promote GH release. It does this by increasing calcium in the cell, which signals the cell to release more GH.

3. GHRP2 + GHRH
The combination of GHRP2 and GHRH results in a much higher GH response than just one or the other. Let me explain GHRH real quick. In short, GHRH (growth hormone-releasing hormone) binds to GHRH receptors to trigger secretion of GH. Synthetic GHRHs replicate this and include; CJC ND, Sermorelin, etc. The most common combination here is GHRP2 + CJC ND. The exact mechanism of synergy is unknown, but my personal hypothesis is that GHRP2 may improve GH efficiency by increasing GHR (growth hormone receptor; prompts the liver to release IGF1 after binding with GH) concentration along with its effect of somatostatin (the hormone that inhibits GH) inhibition. I came to the conclusion that it may increase GHR due to it being shown to increase GHBP (growth hormone binding protein; binds to GH to increase time in bloodstream), which would be connected because GHRs “shed” GHBPs, so a higher GHR count could correlate with more GHBP. This is merely a correlation and should not be taken as causation. It’s a hypothesis and nothing else. GHRP2 also inhibits sst-1 and sst-2 mRNA expression, which basically just means it inhibits somatostatin, obviously helping GHRH produce more GH.

TDLR: GHRP2 + GHRH (like CJC ND) has a much stronger GH response than one by itself. The mechanism is unknown. I hypothesize this is due to an inferred, correlational increase in receptors for GH because there is more binding proteins for GH (which are byproducts of receptor shedding, so more receptors = more binding proteins). Also due to GHRP2 inhibiting somatostatin, the hormone that inhibits GH.

4. Effects on ACTH, Cortisol, and Prolactin
GHRP2 was found to have less prolactin response than TRH (thyrotropin-releasing hormone), which is a hormone that signals prolactin production as a major stimulator. This is good, but you’d still want Cabergoline and/or P5P on hand if your prolactin gets high. GHRP2 was found to have similar ACTH (adrenocorticotropic hormone) and cortisol response as hCRH (human corticotropin-releasing hormone, a peptide that increases both ACTH and cortisol). ACTH signals the adrenaline gland to produce cortisol, and we all know what cortisol does to growth, so this is bad, but easy to mitigate. Take some Ashwagandha, Magnesium, L-Theanine, Omega 3s, and make some lifestyle adjustments for lower cortisol. You should be fine.

TLDR: GHRP2 produces less prolactin than TRH (a main driver of prolactin). Keep Cabergoline and/or P5P on hand in case of elevated prolactin. GHRP2 produces similar amounts of ACTH (stimulates cortisol through adrenal glands) and cortisol as hRCH, a peptide that increases ACTH and cortisol.
